Galanin neurons in the medial preoptic area govern parental behaviour

چکیده
Mice display robust, stereotyped behaviours towards pups: virgin males typically attack pups, whereas virgin females and sexually experiencedmales and females display parental care. Here we show that virginmales genetically impaired in vomeronasal sensing do not attack pups and are parental. Furthermore, we uncover a subset of galanin-expressing neurons in the medial preoptic area (MPOA) that are specifically activated during male and female parenting, and a different subpopulation that is activated during mating. Genetic ablation of MPOA galanin neurons results in marked impairment of parental responses inmales and females and affectsmalemating. Optogenetic activation of these neurons invirginmales suppresses inter-maleandpup-directedaggressionand inducespupgrooming. Thus,MPOAgalaninneurons emerge as an essential regulatory node ofmale and female parenting behaviour and other social responses. These results provide an entry point to a circuit-level dissection of parental behaviour and its modulation by social experience.

Functional, anatomical, and neurochemical differentiation of medial preoptic area subregions in relation to maternal behavior in the mouse.
In rodents, previous findings indicate critical involvement of the medial preoptic area (MPOA) in the neural control of maternal behavior. However, the specification of the particular MPOA subregions involved in maternal behavior and the identification of the neurochemical phenotype(s) of the essential neurons demands additional study. Therefore, we investigated the chemical neuroanatomy of the...
متن کاملGating of the dorsal penile nerve inputs by norepinephrine at the medial preoptic area in rats.
The medial preoptic area neurons related to male sexual behaviour in rats were identified by their responses to dorsal penile nerve stimulation. These neurons were further tested with norepinephrine applied iontophoretically. From the 21 medial preoptic area neurons recorded in urethane anaesthetized rats, 17 neurons responded to dorsal penile nerve stimulation. Excitatory and inhibitory respon...
متن کاملDistribution of galanin-like peptide in the rat brain.
Galanin-like peptide (GALP) is a novel galanin-like peptide isolated from the porcine hypothalamus. To determine the distribution of GALP in the rat brain, we performed immunohistochemical studies using a monoclonal antibody toward the N-terminal sequence of GALP. GALP-immunoreactive neuronal cell bodies were observed only in the arcuate nucleus (Arc), which was further confirmed by in situ hyb...
متن کاملIntermediary role of kisspeptin in the stimulation of gonadotropin-releasing hormone neurons by estrogen in the preoptic area of sheep brain
Introduction: The role of estrogen in the stimulation of gonadotropin-releasing hormone (GnRH) neurons is clear. These neurons do not express estrogen alpha receptors, so other mediator neurons should be present to transmit the positive feedback effect of estrogen to the GnRH neurons. Kisspeptin neurons have an important role in the stimulation of GnRH neurons, so they can be the mediator of...
متن کاملNeurons of origin and fiber trajectory of amygdalofugal projections to the medial preoptic area in Syrian hamsters.
The amygdaloid neurons of origin and the trajectory of amygdaloid fibers to the medial preoptic area of the adult male Syrian hamster were identified by using horseradish peroxidase (HRP) histochemistry. After iontophoresis of HRP into the medial preoptic area, retrogradely labeled amygdaloid neurons were located in the dorsal and caudal parts of the medial amygdaloid nucleus and throughout the...
